【Abstract】 objective to observe the inflammation and immunological changes of Hepatocellular Carcinoma (HCC) in co-culture with hepatic stellate cells (SMMC7721) induced by TGF-1. METHODS: Human Hepatic stellate cells (LX-2) were cultured in vitro and treated with 10 g/l TGF-1 for 24 h. SMMC7721 cells were cocultured with activated LX-2 supernatant for 12 h and treated with Disitetide, flow cytometry observed the expression of CD4 + CD25 + t cells before and after intervention, and the expression of IL-8 and IL-10 before and after intervention by Elisa. Results: After LX-2 was stimulated by TGF-1 at 10ug/l, the expression of-SMA gene and protein level were significantly increased. After co-culture of activated LX-2 supernatant with SMMC7721, the expression ratio of CD4 + CD25 + was significantly higher than that of SMMC7721(p = 0.003) , and the expression level of IL-8 and IL-10 was also significantly higher than that of SMMC7721(p = 0.000) , disitetide inhibited the expression of CD4 + CD25 + after TGF-1 significantly (p = 0.025) , and the expression of IL-8 and IL-10 decreased significantly (p = 0.000) . Conclusion: TGF-1 can induce the activation of HSC and promote the inflammation and immune response of HSC.
